If you want serious power I highly recommend upgrading your turbo along with additional fueling. Since you have access to e85 I would skip meth and go straight to a port injection kit. Without additional fueling you'll be limited to around 16-17psi to redline with the pure stage 2 on pump gas or low ethanol mix. To really take advantage of the upgraded turbo you'll need port injection, upgraded lpfp and an n20 tmap which will allow you to run upwards of 25psi to redline.
To answer your last question, yes you could do the upgrades in stages and add the port injection down the road. Tuning wise you'll be fine with just the jb4 but at these power levels I would recommend a back end flash.
